Lithium-mediate influence on stem cells in vitro culture in neonate rat in anaerobic condition

锂对缺氧状态下新生鼠神经干细胞损伤的影响

Abstract: Obiective To establish hypoxia damage model of neural stem cells (NSCs) and investigate the protection of lithium on them through culture NSCs. Methods NSCs were cultured from neonatal mice,immunocytochemistry was used to examine the Nestin expression of NSC,the forth passage cells were divided into control,hypoxia and lithium groups,NSE and GFAP were used to evaluate cell differentiation,cck-8 was used to evaluate cell proliferation,and NSE and GFAP were used to evaluate cell differentiation. Result NSCs under hypoxia 95%N2,5%CO2 for 30 minutes could establish hypoxia damage model in vitro and certain concentration of lithium protected the damages. Conclusion Lithium can protect NSCs under hypoxia and may prevent NSCs against hypoxia injury.

摘要: 目的 培养神经干细胞,制造缺氧模型并探讨锂对缺氧损伤的神经干细胞是否具有保护作用。方法 选取培养第四代的新生SD大鼠神经干细胞进行缺氧,细胞分为缺氧组和不缺氧组,每组内又分为不加锂组,加锂组(1、2、3、4、5mm)。免疫组织化学检测nestin、NSE和GFAP的表达。结果 95%N2和5%CO2混合气体培养细胞30min能成功构建离体神经干细胞缺氧损伤模型,一定浓度的锂能对缺氧损伤的神经干细胞发挥保护作用。结论 锂对缺氧下神经干细胞有保护作用。
