Long-term effects of repeated inhalation of sevoflurane on behavioristics in neonatal rats.

新生期重复吸入七氟烷对大鼠行为学的远期影响

Abstract: Objective To investigate the effects of repeated inhalation of sevoflurane anesthesia in the neonatal period on capability of behavioristics in juvenile and adult period. Methods 7 day old SD rats (n=24) were randomly divided into 3 groups(n=8 each):2.6% sevoflurane group(sev2.6),1.5% sevoflurane group (sev1.5) and control group (con).The rats were exposed to 2.6% sevoflurane or 1.5% sevoflurane in carrier gas (sev2.6,sev1.5) or simply carrier gas (1 L/min of O2 and 1L/min of air) without any inhalational anesthetic (con) for 1 h at 7,14 d and 21 d after birth (P7,P14,P21).The cued trials was performed at P28 and P29,while the place trials and probe trials were performed in P32~P36 and P91~P95. Results 1)In the juvenile place trials,the escape latency of group sev2.6 and sev1.5 were longer than group con (P<0.05),and the escape latency of group sev1.5 were significantly longer than the group con at P32,P36 (P<0.01).The escape latency of group sev2.6 was longer than sev1.5 (P<0.05).In the adult place trials,the escape latency of sev1.5 were longer than the group con (P<0.05).2)In the adult probe trials,times of crossing target area in sev1.5 were shorter than group con (P<0.05). Conclusion Intermittent repeat inhalation of sevoflurane anesthesia during the neonatal period may injured the declarative memory in juvenile period,and exist concentration dependence.1.5% of sevoflurane on declarative memory function damage may show two peak period,and extended to adulthood,while injured the space association ability and recall inquisition ability in adulthood.

摘要: 目的 探讨新生期重复吸入七氟烷对大鼠幼年期、成年期的行为学影响。方法 7日龄SD大鼠(n=24),随机分为2.6%七氟烷组(n=8,sev2.6组)、1.5%七氟烷组(n=8,sev1.5组)、对照组(n=8,con组)。分别于出生7、14、21 d(P7,P14,P21)吸入2.6%、1.5%七氟烷及运载气体1 h,于P28、P29行可视平台实验, P32~P36及P91~P95行定位航行和空间探索实验。结果 1)定位航行实验逃避潜伏期 幼年期:sev2.6组和sev1.5组较con组延长(P<0.05),其中sev1.5组于P32、P36显著延长(P<0.01); sev2.6组较sev1.5组延长(P<0.05)。成年期:sev1.5组较con组延长(P<0.05)。2)空间探索实验成年期sev1.5组平台区进入次数较con组减少(P<0.05)。结论 新生期重复吸入七氟烷损害幼年大鼠的陈述性记忆功能,且有浓度依赖性。1.5%七氟烷陈述性记忆的损害有两个峰值,可延长至成年,并损害成年期空间联想和回忆探究能力。
