Effect of Quercetin on proliferation and invasion of colon carcinoma LOVO cells and the expression level of carcinoembryonic antigen

Abstract: AIM: To investigate the effect of Quercetin on proliferation and invasion of colon carcinoma LOVO cells as well as the expression level of carcinoembryonic antigen. METHODS: LOVO cells were treated with Quercetin at various concentrations,then determined for proliferation level by MTT assay,for cell adhesion and invasion of LOVO cell by tumor cell adhere to the vascular endothelial cell assay and transwell chamber invasion assay, and for the expression level of CEA by Immunocellulerchemistry (ICC), Western Blot and RT-PCR.RESULTS: Significantly, Quercetin showed dose-dependent inhibitory effect on proliferation of LOVO cells with an IC50 of about 40 μmol/L, also improved the ability of LOVO cells adhere to the vascular endothelial cells and reduced the invasion ability of LOVO cells. Meanwhile, Quercetin significantly inhibited the protein and mRNA expression levels of CEA in LOVO cells.CONCLUSION: Quercetin has inhibition effect on the proliferation invasion ability of LOVO cells,also inhibit the protein and mRNA expression levels of CEA in LOVO cells.
