应用基因芯片研究盐酸小檗碱对大鼠肝脏肿瘤及免疫相关基因表达的影响

Tumor associated and immune associated gene differential expression by cDNA microarray in rat liver treated with berberine chloride

摘要: 目的 研究小檗碱对大鼠肝脏肿瘤及免疫相关基因表达谱的影响。方法 大鼠予以盐酸小檗碱灌胃6 d, 摘取肝脏提取总RNA。用Cy3 和Cy5 两种荧光物质分别标记阴性对照组和盐酸小檗碱组肝组织mRNA, 制备成探针与表达谱芯片进行杂交, 获得的荧光信号图像用计算机扫描后进行数据分析, 计算每点的Cy5 和Cy3 信号强度比值。结果 在被检测的4 096 个基因中, 盐酸小檗碱组差异表达基因236 个, 与肿瘤及免疫相关的差异表达基因共22 个, 其中表达上调的相关基因9 个, 包括细胞色素P450 17 、KAI1 基因、结合蛋白/细胞粘附样分子(Opcml) 、白介素6 、肾特异性蛋白、白介素2 受体、α2-巨球蛋白等。表达下调的相关基因13 个, 包括细胞色素P450 2E1 、睾丸细胞粘附分子1 、S-腺苷-L-甲硫氨酸脱羧酶、去唾液酸糖蛋白受体、肿瘤蛋白翻译调节因子1(Tpt1) 、特异性细胞毒性靶抗原、S100 钙结合蛋白A9 等。结论 小檗碱的抗肿瘤和免疫调节作用可能与一系列肿瘤及免疫相关基因的表达变化有关。

Abstract: AIM: To study the effects of berberine chloride(Ber) on tumor associated and immune associated gene expression profile in rat liver.METHODS: After orally treated with berberine chloride for 6 days, the rats were sacrificed and the livers were taken out to extract total RNA.mRNA from negative control grouPand Ber grouPwere labeled with Cy3-dCTPand Cy5-dCTPrespectively.These probes were hybridized with cDNA microarray.The acquired image was scanned and analyzed by GenePix Pro 3.0 software.The intensity ratio of Cy5 to Cy3 about each spot was calculated.RESULTS: Among 4096 genes, 236 genes exhibited differential expressions in grouPtreated with Ber.22 tumor associated and immune associated genes were differentially expressed with 9 genes up-regulated and 13 genes down-regulated.The genes up-regulated included CYP17, kangai 1, opioidbinding protein/cell adhesion molecule-like (Opcml) , interleukin 6, kidney-specific protein, interleukin 2 receptor and alpha-2-macroglobulin.The genes down-regulated included cytochrome P450 2E1, testicular cell adhesion molecule,S-adenosyl-L-methionine decarboxylase , asialoglycoprotein receptor, tumor protein translationally-controlled 1 (Tpt1) , CTL target antigen and S100 calcium-binding protein A9.CONCLUSION: The anti-cancer effects and immunoregulatory function of Ber may be related with differential expressions of some tumor associated and immune associated genes.
