胰岛素注射方式对2型糖尿病KK小鼠血脂的影响

Effects of insulin treatment on blood lipid in type 2 diabetic KK mice

摘要: 目的:观察胰岛素腹腔和皮下给药对2型糖尿病模型(KK小鼠)血脂的影响。方法: 选择空腹血糖高于20 mmol/mL 的KK小鼠作为2型糖尿病模型,分为腹腔给药组(腹腔给药组, n=9)和皮下给药组(皮下给药组, n=9)。C57BL 小鼠作为正常对照(正常对照组, n=10)。2型糖尿病KK小鼠连续给予高糖高脂
饮食4周,同时IP组每日一次注射胰岛素0.3 mL, 皮下给药组每日一次注射胰岛素0.4 mL(速效胰岛素︰精蛋白锌胰岛素=2︰1), 以维持空腹血糖正常(6.0±1.5) mmol/L。结果: 胰岛素治疗2~4周, 腹腔胰岛素给药可使2型糖尿病KK小鼠血清TG、TC、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C) 的浓度达到正常水平。皮下胰岛素给药只能使TC达到正常水平。结论:与胰岛素皮下给药相比,腹腔胰岛素给药能够更有效地纠正2型糖尿病KK小鼠脂代谢异常。

Abstract: AIM: To study the effect of intraperitoneal and subcutaneous insulin administration on serum lipids in a type 2 diabetic animal model (KK mice).METHODS: KK mice with fasting blood sugar level higher than 20 mmol/mL were set as type 2 diabetic model and divided into subcutaneous injection group(s.c. group, n=9) and introperitoneal injection group(i.p. group, n=9). C57BL mice were set as normal control(C group, n=10). The type 2 diabetic KK mice were given high glucose and fatty for 4 weeks. Meanwhile, insulin was administrated at doses of 0.4 mL/d for s.c. group and 0.3 mL/d for i.p. group to normalize fasting blood glucose concentration(6.0 mmol/L±1.5 mmol/L).RESULTS:After the diabetic KK mice were treated with insulin for 2-4 weeks, the serum TG, TC, LDL-C and HDL-C were recovered to the normal scope by intraperitoneal injections of insulin, but TG, LDL-C and HDL-C were not recovered to the normal level by subcutaneous injections of insulin.CONCLUSION: This result indicates that compared with subcutaneous insulin administration, intraperitoneal insulin administration is more efficacious to correct the disorder of lipid metabolism.
