Nordosin enhanced inhibition of FK506 on the secretion of IL-2 by lymphocyte derived from spleen of mice in vitro

Abstract: AIM: To investigate the effect of Nordosin in inhibiting proinflammatory cytokine with FK506 in vitro and potential applied value in the transplantation immunity.METHODS: ConA-stimulated T-cell proliferation model was made.Then, the expression of IL-2 both in protein and mRNA lever were investigated by ELISA and RT-PCR.Furthermore, the expression of IL-2 receptor, CD25+ on surface of T lymphocyte was also measured by FCM.RESULTS: Compare with those by FK506 use along, Nordosin enhanced the inhibition of FK506 on the expression of IL-2 significantly (P<0.01).The expression of IL-2 mRNA and its receptor CD25+ were also inhibited by their combining medication (P<0.01).CONCLUSION: Nordosin could enhance the inhibition of FK506 on IL-2 secretion significantly, Nordosin maybe become an immunodepression assistant medication to enlarge the pharmacological effect of FK506.
