Protection of CpG oligodeoxynucleotide on human rotavirus-induced diarrhea in suckling mouse model

Abstract: AIM: To explore the protectionmechanism of CpG oligodeoxynucleotide (CpG ODN) on Human rotavirus diarrhea in suckling mice.METHODS: 64 ICR suckling mice were randomly divided into control group (A), HRV infection group (B), CpG ODN pretreatment group (C) and CpG ODN treatment group (D).The treatment response was observed and RV antigen in animal stools was detected.The expressions of Toll-like receptor 9 (TLR9) and interferon-gamma (IFN-γ) in the intestinal tissue were measured with immunohistochemistry.RESULTS: The duration of diarrhea, RV excretion time and the incidence of severe diarrhea of group B were higher than those in group C and group D (P<0.05).The expressions of TLR9, IFN-γ of group C and group D were higher than those of group A and group B (P<0.01), there were a positive correlation in the expression of TLR9 and IFN-γ of group C and group D (P<0.05).CONCLUSION: CpG ODN pretreatment and treatment can improve the expression of TLR9 and IFN-γin intestinal epithelium, CpG ODN has protection to HRV infected suckling mouse.
