Screening of immuno-regulatory components in extract of Yu-ping-feng powder based on delayed type hypersensitivity and Th cytokines

Abstract: AIM: To screen immunoregulatory components by evaluating the effects of different extracts of Yu-ping-feng powder on allergic contact dermatitis (ACD)which is a well known animal model for delayed type hypersensitivity(DTH).Proliferation of T lymphocytes and secretion of the cytokines induced by ConA were also applied to evaluate the effects of extracts of Yu-ping-feng powder.METHODS: Mice ACD were induced by DNCB and the proliferation of T lymphocytes was measured by MTT.The levels of cytokines were determined by ELISA.RESULTS: The swelling degree of mice auricle was reduced significantly by the alcohol extract substance.The T cell proliferation and IFN-γ IL-4 secretion stimulated by ConA were inhibited significantly by this site.Significant effects were also found in the acetic ether extract parts treated groups including reduction of DTH in vivo, inhibition of IFN-γproduction and increase of IL-4 level in vitro.The ligarine extract part was found to inhibit IFN-γand IL-4 secretion in vitro while no significant effect was detected in mice model.As far as the IFN- γ IL-4 ratio was concerned, the alcohol extract part, the ligarine extracting site and the acetic ether extracting site had certain inhibitory action.CONCLUSION: The active components for DTH are wherein the alcohol extracting site and the acetic ether extracting site of Yu-ping-feng powder and their effects were related to the regulation of Th1 Th2 balance.At the same time the results indicate that it is reasonable for Yu-ping-feng used as powder in clinic.
