Pharmacokinetics of levobupivacaine for brachial plexus block in pediatric patients

Abstract: AIM: To evaluate the clinical pharmacokinetics of levobupivacaine for brachial plexus block in pediatric patients. METHODS: 9 pediatric patients,undergoing elective upper shoulder surgery,were injected with levobupivacaine 0.375﹪(0.4 mL/kg)for interscalene brachial plexus block.The plasma concentration of levobupivacaine were determined at 10,20,30,45,60,90,120,180,240,360,480 and 840 min after injection,and the pharmacokinetic parameters were calculated with 3P97 software package. RESULTS: The plasma concentration-time curve was fitted to a two-compartment model,and the main pharmacokinetic parameter such as t1/2β,tmax and Cmax was (4.77±0.52),(0.28±0.14) h and (1.02±0.22) mg/L. CONCLUSION: Levobupivacaine 0.375% can be used for brachial plexus block in children safely,and its clinical pharmacokinetics is fitted to a two-compartment model.
