Advances in understanding the role of TGF-β/Smad signalling pathways in the pathogenesis of liver fibrosis

Abstract: Liver fibrosis is the common pathway of virtually various chronic liver diseases to the liver cirrhosis, which is characterized by the excessive accumulation of collagen-based extracellular matrix for imbalance between synthesis and degradation. It is regulated by multiple cell signaling pathways and a series of cellular molecular information networks. Transforming growth factor-β (TGF-β) is one of the strongest profibrotic cytokines, and TGF-β/Smad signaling pathway is the main way of various types of TGF-β signal. It activates hepatic stellate cell (HSC) to promote ECM production. This review is related to the effect of TGF-β/Smad signaling pathway in the progress of liver fibrosis.
