Effects of 5-Fluorouracil on cell proliferation and potassium channel function in oral cancer cell line Tb

Abstract: AIM: To evaluate the effects of 5-Fluorouracil (5-FU) on cell proliferation and potassium channel function in oral cancer cell line Tb.METHODS: After treatment with 5-FU for 24 h, the proliferation of Tb cells was determined by MTT and clone formation assay, the membrane potential and potassium channel current of Tb cells were recorded by whole-cell patch clamp technique.RESULTS: Compared with the control group (CON), 5-FU inhibited proliferation of the Tb cells.At the same time the membrane was depolarized and the potassium channel current significantly decreased compared with the CON group.CONCLUSION: Potassium channel may play an important role in 5-FU-inhibited the proliferation of Tb cells.
