Effects of erycin on NF-κB and Th2 in rats with asthma

Abstract: AIM: To explore the effects of erycin on the expression of cytokines such as NF-κB and Th2 in asthmatic rats. METHODS: The peripheral blood monouclear cells(PBMCs) were separated and cultured from rats in bronchial asthma, normal control and erycin-treatment groups.The concentrations of IL-4, IL-5 and IL-13 in plasma, the expressions of NF-κB mRNA and NF-κB protein in PBMCs were detected by enzyme linked immunosorbent assay (ELISA), reverse transcription polymerase chain reaction (RT-PCR), Western blotting, respectively. RESULTS: Compared with normal group the expressions of NF-κB mRNA and NF-κB protein in PBMCs and the concentrations of IL-4, IL-5 and IL-13 in supernatant were higher in asthmatic group (P<0.05).Compared with the asthmatic group, the above indexes were decreased significantly in erycin-treatment group (P<0.05).There were positive correlations between the expression of NF-κB protein in PBMCs and the concentrations of IL-4, IL-5 and IL-13 in supernatant (r =0.78, 0.86 and 0.71, P<0.05). CONCLUSION: NF-κB may play a role in patho-physiological course of asthma.Erycin could effectively treat asthma by inhibiting the expression of NF-κB.
