    Study on differential expression of the mi-RNA in the deep second degree burns

    AIM:To study the differential expression profile of the miRNA in deep second-degree burns and normal skin tissue, and to investigate the effects of differential expression of the miRNA on wound healing. METHODS: The differential expression of miRNA in deep second-degree burns and normal skin tissue were screened by gene chip detection technology, and the expression of some miRNA was verified by RT-PCR. Target gene prediction was carried out through gene databases TargetScan, miRanda and mirbase. The targeting function analysis and signaling pathway analysis for the predicted target genes were completed with the help of GO-analysis.RESULTS:Compared with the normal group, there were 41 miRNAs genes up-regulated and 124 miRNAs genes down-regulated in the deep second-degree group, and greater differences were shown in some genes, including miR-210-3P, miR-99a-5P, miR-99b-3P, miR-99b-5P, miR-143-5P, miR-29c-5P, miR-15b-5P, miR-323a-3P, miR-424-5P, miR-125b-1-3P, miR-125a-3P, miRNA-184, which were involved in the inflammatory response, immune response and the healing process. CONCLUSION:The regulatory mechanism of miRNA with differential expression and its corresponding target genes in the healing of burn wounds was confirmed at the gene level, which provided targets for the precise treatment for burn wounds.

    Effects of NBP on energy metabolism of human brain microvascular endothelial cells under the condition of glucose deprivation

    AIM: To study the effect and protective effect of butylphthalide (NBP) on the energy metabolism of human cerebral microvascular endothelial cells (HBMEC) under the condition of glucose deprivation (OGD).  METHODS: HBMEC cells were treated by OGD. The cells were divided into control group, model group, low dose group, middle dose group and high dose group. The control group was normal cultured, the model group were OGD treated, the low dose group received OGD+5 mol/L NBP, the middle dose group received OGD+10 mol/L NBP, and the high dose group received OGD+20 mol/L NBP. The CCK-8 kit was used to detect the cell viability.The leaking rate of lactate dehydrogenase (LDH), sodium potassium ATPase (Na+-K+-ATPase), calcium triphosphatase (Ca2+-ATPase) activity and intracellular calcium concentration were detected acording to the description of kit,respectively. The flow cytometry was used to detect the apoptosis rate and Hoechst33342 staining. The high performance liquid chromatography was used to detect the contents of ATP, ADP and AMP in the cells, and the EC value was measured.RESULTS:The cell activity, Na+-K+-ATPase and Ca2+-ATPase in the model group was decreased, but the intracellular calcium concentration, the rate of apoptosis and the leakage rate of LDH were significantly increased. After NBP intervention, the activity of cell, the activity of Na+-K+-ATPase and Ca2+-ATPase, the concentration of calcium ion in the cell and the rate of cell apoptosis were reduced. The content of ATP, ADP and EC value in the cells was increased while the content of AMP was decreased in a dose-dependent manner. CONCLUSION: NBP can protect HBMEC injury under OGD, and its mechanism is related to improving energy metabolism and inhibiting intracellular calcium.

    Protective effects of Ferulic acid on acute liver injury induced by D-GalN/LPS in mice

    AIM:To study the protective effects of Ferulic acid on acute liver injury induced by D-GalN/LPS in mice. METHODS:A total of 60 BALB/c mice were divided into six groups:control group,D-galactosamine/lipopolysaccharide(D-GalN/LPS) model group(600 mg·30 μg/kg),bifendate group(200 mg/kg),the high,middle and low dose groups of Ferulic acid (30, 15, 7.5 mg/kg, respectively), with 10 mice in each group. All groups received intragastric administration once a day for 21 days. Mice were sacrificed 2 h after the last administration to collect liver tissue and blood from eyeball, and ALT, AST, SOD, GSH-Px, MDA were detected. The pathological changes of liver tissue were observed, and liver index was calculated according to the body weight of mice.RESULTS:Compared with D-GalN/LPS model group, bifendate group(200 mg/kg) and the high, middle and low dose groups of Ferulic acid presented siginificant decrease of the liver index, serum ALT, AST and MDA (P<0.05), and significant increase of SOD and GSH-Px (P<0.05). Pathological analysis of the liver tissue showed that bifendate and different doses of Ferulic acid could alleviate the hepatocytosis and necrosis. CONCLUSION: Ferulic acid has protective effect on acute liver injury induced by D-GalN/LPS in mice. The mechanism maybe related to the anti-oxidation effect of Ferulic acid.

    Effects of angelica sinensis polysaccharide on TLR4/MyD88/NF-κB pathway in rats with diabetic neuropathy

    AIM: To analyze the effects of angelica sinensis polysaccharides on TLR4/MyD88/NF-κB signaling pathway in the inhibition of diabetic neuropathy (DPN) in rats. METHODS: A total of 105 SPF-level Wistar male rats were randomly selected and 15 were selected as the normal group, and the remaining 90 rats were used to prepare the DPN model. Seventy-five models were successfully established and were randomly divided into 5 groups, i.e. the model group, the angelica polysaccharide low, medium and high dose groups and positive control group, with 15 rats in each group. The rats in the model group and the normal group were intragastrically administered with 2 mL/100 g normal saline. The rats in the low, middle and high doses of angelica polysaccharide were given the polysaccharide of angelica sinensis polysaccharide, and the positive control group was given the Mi Kebao + metformin solution. The administration volume was 2 mL/100 g once a day for a continuous gavage of 10 days. After 3 and 10 days of administration, the mechanical contraction threshold (PWT) and heat-shrinking reflex latency (PWL) were measured. After the last administration, the rats were tested for fasting blood glucose, serum malondialdehyde (MDA) and superoxide dismutase. (SOD), catalase (CAT) activity, glutathione peroxidase (GSH-Px), interleukin-6 (IL-6), myelin basic protein (MBP), TNF-α And C-reactive protein (CRP) content, detecting motor motor conduction velocity (MNCV), sensory nerve conduction velocity (SNCV), rat sciatic nerve factor-κB (NF-κB) mRNA, myeloid molecular factor 88 (MyD88) mRNA and Toll-like receptor 4 (TLR4) mRNA expression. RESULTS: The fasting blood glucose and MDA content in the model group and the low dose group of angelica polysaccharide were significantly higher than those in the normal group. The contents of CAT, SOD and GSH-Px were significantly lower than those in the normal group. The fasting blood glucose and MDA content in the middle and high dose groups of angelica polysaccharide were significant. The levels of CAT, SOD and GSH-Px in the model group and the positive control group were significantly higher than those in the model group and the positive control group, and the difference was statistically significant (P<0.05). The levels of serum IL-6, MBP, TNF-α and CRP in the model group and angelica polysaccharide low dose group were significantly higher than those in the normal group, and the serum IL-6, MBP and TNF-α in the middle and high dose groups of angelica sinensis polysaccharide. The CRP content was significantly lower than that of the model group and the positive control group, and the difference was statistically significant (P<0.05).The expressions of NF-κB mRNA, MyD88 mRNA and TLR4 mRNA in the model group and the low dose group of angelica sinensis polysaccharide were significantly higher than those in the normal group, and the expression of NF-κB mRNA, MyD88 mRNA and TLR4 mRNA in the middle and high dose groups of angelica sinensis polysaccharide. Both were significantly lower than the model group and the positive control group, and the difference was statistically significant (P<0.05). CONCLUSION: Angelica sinensis polysaccharides can significantly reduce the serum inflammatory factors and oxidative stress response in DPN rats. The mechanism may be that it inhibits the expression of TLR4, which affects the downstream gene expression of MyD88 signaling pathway, which is related to the inhibition of NF-κB activation.

    AIM: To observe the effects of endoplasmic reticulum associated degradation protein Derlin-1 on the apoptosis of human colon cancer cells (SW480) and its possible mechanism.  METHODS: The qualified recombinant plasmid sh-Derlin-1-pGPU6/Neo was transfected into SW480 cells with Derlin-1 knocking down. The expression level of Derlin-1 was detected at both mRNA and protein levels by real-time PCR and Western blot. CCK8 assay and flow cytometry were performed to detect the proliferation and apoptosis of SW480 cells. We examined the expression of apoptosis associated proteins (Bcl-2 and Bax) and ERK phosphorylation followed by Derlin-1 low-expression by Western blot. RESULTS: The Derlin-1 knocking down plasmid was constructed successfully. SW480 cells with Derlin-1 knocking down had been established successfully. Compared with control group, the proliferation rates of sh-Derlin-1 SW480 cells were significantly decreased, while the apoptotic rates of sh-Derlin-1 SW480 cells were significantly increased (P<0.05); the expression levels of Bcl-2 and p-ERK in sh-Derlin-1 group were significantly decreased, while the expression level of Bax was greatly increased (P<0.05). CONCLUSION: The decreased level of Derlin-1 can suppress the proliferation and induce the apoptosis of SW480 cells through suppressing the activity of ERK signaling pathway.

    Blood concentration monitoring of hydroxychloroquine in patients with autoimmune diseases

    AIM: To explore the factors that influence the blood concentration of hydroxychloroquine (HCQ) including dosage, treatment course, and combination of the drug (hormone, cyclophosphamide, mycophenolate mofetil and methotrexate), as well as the disease type, age and gender of the patients. METHODS: During July 2017 to June 2018, blood samples were collected from autoimmune diseases patients who had been treated with HCQ for more than 6 months in department of rheumatology and immunology of our hospital. The blood concentration of HCQ was monitored by high performance liquid chromatography (HPLC). Statistical analyses were performed by SPSS 16.0. RESULTS: The blood concentration of HCQ was relatively high in patients treated with 0.4 g HCQ per day. Patients that over 50 years old and/or have been treated with HCQ for more than 10 years presented significant higher HCQ blood concentration (P<0.05). Other factors including disease type, gender and drug combination didn't obviously affect the concentration of HCQ. Moreover, we found that the incidence of adverse drug reactions was increased along with the increase of HCQ blood concentration. CONCLUSION: The blood concentration of HCQ is mainly affected by the dosage, the age of the patients, and the course of HCQ treatment. The equivalent therapeutic effect can be achieved with lower HCQ dosage in elderly and long-term treatment patients. Adjustment the dosage according to the blood HCQ concentration monitoring could decrease the incidence of adverse drug reactions.

    AIM: To investigate the genetic polymorphism of CYP2C19 among patients requiring percutaneous coronary intervention(PCI), and to determine the anticoagulation therapy according to genotypes so as to observe its clinical efficacy. METHODS: CYP2C19 variants were detected by PCR-RFLP method in patients requiring PCI. The extensive metabolism(EM)group was treated with clopidogrel with loading dose of 300 mg at the first time and 75 mg/d for 12 months.While the intermediate metabolism (IM) group was treated with clopidogrel with loading dose of 300 mg at the first time and 150 mg/d for 12 months.The poor metabolism (PM) group was treated with ticagrelor with loading dose of 180 mg at the first time and 90 mg,bid for 12 months.All three groups were given Aspirin of 100 mg/d. During the follow-up of 12 months,the incidence rates of major adverse cardiac events (MACE) in the three groups were observed and compared. RESULTS: Five genotypes of CYP2C19 were found in these subjects,and that the frequency of EM was 37.6%,IM 55.6% and PM 6.8%.No significant difference related to the incidence rates of major adverse cardiac events in the three groups during the follow-up of 12 months was observed. CONCLUSION: CYP2C19 genotype can instruct the medication of anticoagulation therapy; patients with fast metabolism can receive regular dose of clopidogrel, while patients with mutation should consider larger dose of clopidogrel or other anticoagulation drugs.

    Effects of CYP2A6*4 gene polymorphism on dexmedetomidine pharmacokinetics

    AIM: To determine the dexmedetomidine pharmacokinetics of CYP2A6*4 allele in Chinese patients with high mutation frequency of CYP2A6*4 in order to provide clinical references. METHODS: Thirty-one surgery patients received 0.5 μg/kg dexmedetomidine via intravenous pump. Their plasma concentrations at multiple time-points and polymorphism of CYP2A6*4 were determined and statistically analyzed.RESULTS:Nine patients were *1/*4 or *4/*4 , and 22 patients were *1/*1. The main pharmacokinetic parameters were area under curve (AUC) (1 396.19±332.47)h·ng·L-1, peak blood concentration (Cmax) (495.50±104.90)ng/L, distribution volume (V) (0.68±0.20)L/kg, clearance (CL) (0.38±0.11)L·h-1·kg-1, distribution half-life (t1/2α) (0.05±0.01)h, elimination half-life (t1/2β) (2.53±0.04)h. No significant pharmacokinetic differences were found among CYP2A6*1/*1, *1/*4, and *4/*4 patients.CONCLUSION: In Chinese patients treated with dexmedetomidine, t1/2β is consistent with that published, but t1/2α, V and CL are lower. It is unnecessary to consider the mutation when developing the precision regimen of dexmedetomidine.

    AIM: To analyze the predictive prognostic value of the gross tumor volume (GTV) before chemo-radiotherapy and the tumor volume reduction rate (VRR) after concurrent chemo-radiotherapy (based on PF program) in patients with non-surgical esophageal cancer. METHODS: Clinical data from 77 patients with non-surgical esophageal cancer who under the concurrent chemo-radiotherapy from June 2010 to October 2013 at the First Affiliated Hospital of Fujian Medical University were retrospectively analyzed. Of the 77 patients, 21(27.3%) were at stage T2, 37 (48.0%) at stage T3, and 19 (24.7%) at stageT4; clinical stage II (34, 44.1%), stage III (28, 36.4%), stage IV (15, 19.5%). The tumor length, GTV, and VRR ((pGTV-iGTV)/pGTV) were measured according to CT images before and at the end of chemo-radiotherapy. The Kaplan-Meier method was used to analyze the survival rate. The COX risk regression model was used to analyze the independent predictive value of GTV, VRR, age, gender, T stage, clinical stage, tumor length, pathological type, RT dose and smoking history in the survival rate of non-surgical esophageal cancer who under the concurrent chemo- radiotherapy. RESULTS: The follow-up time ended on September 1st, 2018. The follow-up rate was 94.8% (73/77), of the 73 cases, 64 cases died, and the median survival time was 15 months of the whole group: the 1-year, 3-year and 5-year survival rates were 65.8%, 24.7%,14.8%, respectively; GTV≥25 mL group: of the 35 cases, 32 cases died, the median survival time was 10 months, the 1-year, 3-year and 5-year survival rates were 42.9%,11.4%,8.6%, respectively; GTV<25 mL group: of the 38 cases, 32 deaths. The median survival time was 27 months, and the 1-year, 3-year and 5-year survival rates were 68.4%, 36.8%, 20.3%, respectively; the difference was statistically significant (χ2=9.542, P=0.002); VRR≥0.3 group: of the 41 cases, 33 cases died, the median survival time was 16 months, the 1-year, 3-year and 5-year survival rates were 75.6%, 29.3%,19.5%, respectively. VRR<0.3 group: of the 32 cases, 31 cases died, and the median survival time was 12 months. the 1-year, 3-year and 5-year survival rates were 62.5%,18.8%,7.8%, respectively, and the difference was not statistically significant (χ2=2.419,P=0.120). Univariate analysis by Log-Rank showed: GTV≥25, (HR=2.136, P=0.003), which was related to the prognosis of patients; Multivariate Cox regression analysis showed that VRR(HR=0.309, P=0.001),GTV (HR=6.557, P=0.000) had an effect on survival and were independent prognostic factors. CONCLUSION: The gross tumor volume (GTV) before chemo-radiotherapy and the tumor volume reduction rate (VRR) are independent factors affecting prognosis. It is recommended to adjust the comprehensive treatment plan according to the GTV and VRR.

    Clinical study of lipid type artificial tear in cataract patients with postoperative dry eye by phacoemulsification

    AIM: To investigate the clinical effect of lipid artificial tears on patients with dry eye after phacoemulsification.  METHODS: A total of 90 cases of dry eyes after cataract surgery were selected after phacoemulsification in our hospital from March 2015 to May 2017 and they were divided into observation group (45 cases) and control group (45 cases) according to the random number table method, the control group used sodium hyaluronate eye drops, the observation group used lipid type artificial tears (gel carbomer), the two groups were tested for ocular surface disease index (OSDI), break-up time (BUT), corneal fluorescein staining (CFS) and schirmer I test (SIT) at preoperative, one week after operation, one month after operation and three months after operation. RESULTS:The total effective rate of observation group (88.1%) was significantly higher than that (65.0%) of control group (P<0.05); one week after operation, there was no statistically significant difference in OSDI scores between the two groups (P>0.05); one month and three months after operation, OSDI scores in both groups were significantly lower than one week after operation (P<0.05), and the OSDI score of observation group was significantly lower than that of control group (P<0.05); one week, one month and three months after operation, the BUT of observation group were significantly higher than those of control group (P<0.05); one week, one month and three months after operation, the CFS scores of observation group were significantly lower than those of control group (P<0.05); one week after operation, there was no statistically significant difference in SIT experiment results between the two groups (P>0.05); one month and three months after operation, the results of the SIT experiment were significantly better than that of control group (P<0.05). CONCLUSION: Lipid-type artificial tear is effective in treating patients with dry eye after phacoemulsification, and it can obviously alleviate the patient's dry eye symptoms and helps the recovery of tear film, which is worthy of clinical promotion.

    Short-term clinical observation of moderate to severe active rheumatoid arthritis treated with tocilizumab combined with disease modifying antirheumatic drugs

    AIM: To study the clinical efficacy and adverse reactions of tocilizumab combined with disease modifying antirheumatic drugs (DMARDs) in the treatment of moderate-to-severe active rheumatoid arthritis (RA) for 12 weeks. METHODS: Twenty patients with moderate-to-severe active RA were treated with 8 mg/kg of tocilizumab at 0, 4th, and 8th week and treated with DMARDs. The effects and adverse reactions were observed at 4th week, 8th week and 12th week. The observed indicators were erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), rheumatoid factor (RF), hemoglobin, platelet, DAS28, hepatic function, renal function, ACR20, ACR50, overall evaluation of the patient and physician to disease. RESULTS: Compared with baseline, ESR, CRP, RF and platelet counts in 20 RA patients were significantly decreased at 4th week, 8th week, and 12th week, and the difference was statistically significant (P<0.01); DAS28 was significantly improved at 4th week of treatment, and further improved at 8th week of treatment, the difference was statistically significant (P<0.01); and hemoglobin increased significantly at 4th weeks, and further increased at 12th week, the difference was statistically significant (P<0.01). The overall evaluation scores of the patients and doctors decreased continuously, the difference was statistically significant (P<0.01); At 8th week and 12th week, ACR20 reached 90%, 95%, and ACR50 reached 80%, 90%; 1 of 20 patients appeared upper respiratory tract infection, there were no liver, kidney and hypersensitivity reactions. CONCLUSION: Tocilizumab combined with DMARDs in the treatment of RA within a short period of time improve the patient's symptoms, signs and inflammation indicators with no significant adverse reactions occurred.

    Safety and efficacy of Guizhi Fuling capsule in improving postoperative pregnancy outcome in patients with mild endometriosis infertility

    AIM: To explore the safety and efficacy of Guizhi Fuling capsule in improving the pregnancy outcome of patients with mild endometriosis infertility.  METHODS: Sixty cases of laparoscopy confirmed mild endometriosis admitted to our department of gynecology, according to the random control table, they were divided into the control group and the experimental group. Thirty cases in the control group were given the routine Goserelin Acetate Sustained-Release Depot subcutaneous injection, and thirty cases in the experimental group were treated with Guizhi Fuling capsule orally. After treatment, the clinical efficacy, ovulation rate, pregnancy success rate and abortion rate, ERK-5, p-ERK of two groups were compared. RESULTS: Compared with the control group, the total clinical effective rate was higher in the experimental group (P<0.05), and the ovulation rate (90%, 27/30) in the experimental group was higher than that in the control group (66.67%, 20/30) (χ2=4.81, P<0.05), and the survival rate (76.67%, 23/30) in the experimental group was higher than that in the control group (33.33%, 10/30) (χ2=11.38, P<0.05). In the experimental group, the abortion rate (3.33%, 1/30) was lower than that in the control group (20%, 6/30) (χ2=4.04, P<0.05). After treatment, the ERK-5 levels in the experimental group and the control group were (10.81±3.29), (13.34±2.16) μg/mL, respectively; p-ERK were (0.99±0.17), (1.59±0.27) μg/mL, respectively (all P<0.05). CONCLUSION: Guizhi Fuling capsule can significantly improve the clinical efficacy and pregnancy success rate of patients with mild endometriosis infertility.

    AIM: To investigate the therapeutic effect of the patients with malignant lymphoma treated with high dose chemotherapy combined with autologous hematopoietic stem cell transplantation (AHSCT).METHODS: The follow up data in 40 patients with malignant lymphoma who recieved AHSCT in Anhui Province Cancer Hospital from March 2013 to February 2018 were analyzed retrospectively.Of the 40 patients, 4 were Hodgkin's lymphoma (HL) and 36 were non-Hodgkin's lymphoma (NHL). To mobilize peripheral hematopoietic stem cell, etoposide combined with G-CSF was used. Preconditioning regimens included BEAM, CBV and BEAC. RESULTS:All patients were collected enough peripheral blood stem cells.None of the patients were died for the risks of AHSCT. The median time of the absolute neutrophil count (ANC) implanted was 10 (7-17) days.And the median time of platelet count implanted was 12 (6-25) days.The median survival time was 15 (1-53) months, progression free survival (PFS) was 72.5%,and overall survival (OS) was 82.5%.The patients who achieved CR status before transplantation could have a favorable prognosis. The prognosis in the patients with IPI score>2 was poor.CONCLUSION:AHSCT can improve the prognosis of patients with malignat lymphoma.

    AIM: To observe the clinical effect of non-invasive cardiac output monitoring (USCOM) combined with milrinone in children with severe pneumonia and heart failure and the effect on brain natriuretic peptide levels. METHODS: Seventy-eight children with severe pneumonia and heart failure were randomly divided into experimental group and control group, with 39 cases in each group. The control group received oxygen, anti-infection, anti-heart failure and maintenance of ionic acid-base balance treatment, and was given intravenous infusion of dopamine 5 μg·kg-1·min-1 for 24 h. The observation group was given milrinone 0.5 μg·kg-1·min-1 based on the control group, intravenous drip, maintained for 24 h. Both groups received 5 d treatment, and the clinical symptoms, signs improvement rate, cardiac function index, inflammatory factors, brain natriuretic peptide levels, and adverse drug reactions were compared between the two groups. RESULTS: After treatment, the irritability disappeared, the wet rales disappeared, the liver retraction and the ruddy rosy improvement rate were higher in the observation group than in the control group (P<0.05). The shortening rate of breast-feeding time in the observation group was lower than that in the control group (P<0.05). After treatment, the HR level in the observation group was lower than that in the control group (P<0.05); the Vpk, FTc and SVI levels in the observation group were higher than those in the observation group. After treatment, the levels of TNF-α, IL-6, IL-8, IL-10 and brain natriuretic peptide in the observation group were lower than those in the control group (P<0.05); No significant difference of nausea and vomiting, diarrhea, blood pressure fluctuation and liver and kidney dysfunction (P>0.05) was observed between the two groups. CONCLUSION: Milrinone can achieve higher short-term curative effect in patients with severe pneumonia and heart failure, which can shorten the clinical symptoms and signs, reduce the incidence of inflammatory factors and adverse drug reactions, and give USCOM monitoring during the treatment to reflect the heart function of patients. Levels help to guide clinical treatment and are worth promoting.

    Effects of Yishen Kangliu recipe in combination with IL-2 peritoneal perfusion in treatment of ovarian cancer-associated ascites

    AIM: To investigate the therapeutic effect of Yishen Kangliu recipe combined with IL-2 peritoneal perfusion on advanced ovarian cancer-related ascites. METHODS: Ninety-eight cases of ovarian cancer-related ascites were divided into experimental group (49 cases) and control group (49 cases) by random number table. Both groups were treated with paclitaxel combined with carboplatin. On the basis of this, the control group was given intraperitoneal infusion of IL-2, while the experimental group IL-2 intraperitoneal perfusion combined with Yishen Kangliu receipe treatment. The effective rate of ascites control, CEA, CA125, HE4, VEGF, T lymphocyte subsets, IgA, IgM, IgG level changes, adverse reactions and Karnofsky score changes, and PFS of the groups were compared. RESULTS: The total effective rate of the experimental group was significantly higher than that of the control group (P<0.05). The total remission rate and control rate of CA125 in the experimental group were 71.4% and 95.9% respectively, which were significantly higher than 46.9% and 83.7% in the control group (P<0.05). After treatment, the levels of CEA, CA125, HE4 and VEGF in the experimental group were significantly lower than those before treatment and after the control group (P<0.05). After treatment, the levels of CD3+, CD4+, CD4+/CD8+, NK cells and IgM, IgG in the experimental group were significantly higher than those before treatment, and the experimental group was significantly higher than the control group (P<0.05). Gastrointestinal reactions, leukopenia, thrombocytopenia, elevated transaminases, elevated urea nitrogen and peripheral neurological symptoms in the experimental group were significantly lower than those in the control group (P<0.05). PFS in the experimental group was 6.0 (3-12) months, which was significantly longer than that in the control group 4.0 (2-10) months (χ2=8.113, P<0.01). CONCLUSION: Yishen Kangliu recipe combined with IL-2 intraperitoneal perfusion in the treatment of ovarian cancer complicated with malignant ascites i can effectively inhibit the proliferation of tumor cells and improve the quality of life of patients with high safety.

    AIM: To investigate the curative effects of Wuling capsule combined with agomelatin in female patients with depression and sleep disorders and its effect on neurotransmitters and endocrine hormones. METHODS: A total of 128 female patients with depression and sleep disorder who were treated in our hospital from January 2016 to May 2018 were selected and were randomly divided into combined group (64 cases) and single group (64 cases). The single group was treated with agomelatine and the combined group was treated with agomelatine and Wuling capsules. After 8 weeks of continuous treatment, the clinical efficacy, HAMD score, PSQI score, neurotransmitter and endocrine hormone levels were observed in the two groups. RESULTS:After 8 weeks of treatment, the total effective rate was 93.75% in the combined group and 81.25% in the single group, the difference between the two groups was statistically significant (P<0.05). After 4 and 8 weeks of treatment, the HAMD score and PSQI score of the two groups were significantly lower than those before treatment (P<0.05), and the combined group was significantly lower than the single group (P<0.05). After treatment, the levels of 5-HT, NE and E2 were significantly higher than those before treatment (P<0.05), and the combined group was significantly higher than the single group (P<0.05). After treatment, FSH levels in the two groups were significantly lower than those before treatment (P<0.05), and the combined group was significantly lower than the single group (P<0.05). There was no significant difference in adverse reactions between the two groups (P>0.05). CONCLUSION: Wuling capsule combined with agomelatin has a good effect on female depression patients with sleep disorders, it can significantly improve the levels of sleep disorders, neurotransmitters and endocrine hormones, and does not increase adverse reactions, which is of high clinical value.

    AIM: To establish the drug use evaluation(DUE) for intravenous injection (IV) administration of voriconazole and to provide reference for rational use of voriconazole. METHODS: According to voriconazole instructions and relative references, DUE criteria for IV administration of voriconazole were established. A total of 121 cases from July 2017 to July 2018 were retrospectively evaluated by the established evaluation and analyzed in respects of medication indication, medication duration, medication results, management indexes, etc. RESULTS: The results for DUE of voriconazole was that the coincidence rate of medication indication was 66.9%(target value was 90%), the rates of administration, solvent selections and preparation methods were all 100%(target value was 100%), the rates of dosage and course of treatment were 31.4% and 81.0% (target value was 95%), microbial inspection rate was 43.8%(target value was 80%),the effective rate of drug use was 50.4% (target value was 80%) and the compliance rate of authority management was 73.6% (target value was 100%). CONCLUSION: Established DUE criteria of IV administration of voriconazole can evaluate and standardize the clinical utilization of voriconazole.

    Angiotensin (1-7) reverse regulation of the classic ACE-AngII-AT axis through the branch of the new role of angiotensin converting enzyme ACE2-Ang (1-7) -Mas receptor axis. Ang (1-7) is widely distributed in the supraoptic and paraventricular nucleus of hypothalamus. It has effects of dilating blood vessels, anti-arrhythmia, anti-inflammatory and so on, which could protect the cardiovascular and central nervous system by affecting the synthesis and releasing other central neurotransmitters through bradykinin, nitric oxide synthase (NOS), cyclooxygenase (COX) and some signaling pathways. This paper reviews the distribution of Ang (1-7) and the diversity of the biological functions in the central nervous system.

    Since the China Food and Drug Administration (CFDA) released the "Announcement on Conducting Self-examination and Verification of Drug Clinical Trials" on July 22, 2015, the integrity and authenticity of clinical trial data has increasingly aroused the attention of sponsors, clinical drug trial institution, and contract research organizations (CROs). In order to further improve the standardization management, quality and efficiency of Phase I clinical trials and ensure the accuracy and reliability of various clinical data, our institute brought in the management system (BEIS) of Hangzhou ECRFPlus Technology Company Ltd. for clinical trials in 2017. Comparative analysis of clinical trials completed with or without using BEIS system showed that adoption of Electronic Management System resulted in many unsurpassed advantages such as less mistakes, fewer queries, lower study cost, and compressed study timeline. Moreover, when BEIS system docking to the third party system such as hospital information management system (HIS), laboratory information management system (LIS) and ECG network information system, it allows for the real-time digitalized information management of the whole process of clinical trials and integration of data capturing, data cleaning, trial management and even data mining in one system, which greatly improved the efficiency and quality of clinical study.
