Establishment of in situ perfused rat intestine-liver model for evaluation on chlorogenic acid metabolism

Abstract: AIM: To establish a model of in situ perfused rat intestine-liver preparation and HPLC-MS method in the determination of chlorogenic acid, caffeic acid,hippuric acid and ferulic acid in perfusate, and investigate chlorogenic acid metabolism in intestine and liver of rats by the model.METHODS: Chlorogenic acid and its metabolites (caffeic acid, ferulic acid and hippuric acid)were detected by HPLC-MS from samples of in situ perfused rat intestine-liver model.RESULTS: Main active metabolite of chlorogenic acid is ferulic acid in in situ perfused rat intestine-liver model after intraduodenal injection to rats, and trace caffeic acid and abundant hippuric acid were also found in perfusate.CONCLUSION: In situ perfused rat intestine-liver models are suitable for the study of metabolism of chlorogenic acid in intestine and liver of rats.Chlorogenic acid was extensively metabolized in rat intestine.
