The role of ginkgo biloba extract on the hepatic GRP78 and CHOP expression in rats with liver fibrosis

Abstract: AIM: To observe the effect of ginkgo biloba extract (GbE) on expression of GRP78 and CHOP in endoplasmic reticulum (ER) stress-related pathway in rats with liver fibrosis, and to explore the mechanism of anti-hepatic fibrosis of GbE. METHODS: The model with liver fibrosis in rats were induced by intraperitoneal injection of 40% CCl4。Meanwhile, all the rats were administrated with different doses of GbE (200, 100 and 50 mg·kg-1·d-1 body weight) by gavage. After the experiment, the mRNA and protein expressions of hepatic GRP78 and CHOP were detected with RT-PCR and immunohistochemical method. RESULTS: The mRNA and protein expression of hepatic GRP78 and CHOP in the rats of the model were more obviously enhanced than in the normal (P<0.05, P<0.01), which were obviously decreased after the treatment with GbE (P<0.05, P<0.01). CONCLUSION: ERS reaction occurs on the liver of rats with liver fibrosis. And that GbE down-regulate the mRNA and protein expressions of hepatic GRP78 and CHOP probably is one of its mechanism of anti-liver fibrosis.
