Abstract: AIM: To study the protective effect of reduced glutathione hormone (GSH) on the dysfunction of diaphragmatic muscle in type 2 diabetic rats and investigate the changes of diaphragm nebulin and titin mRNA expression.METHODS: Male SD rats were randomly divided into control group (CON group), type 2 diabetes mellitus group (DM group) and prototype glutathione treatment group (GSH group). The isolated diaphragm contractile force was measured, diaphragm SOD activity and MDA content, nebulin and titin mRNA levels in diaphragmatic muscle tissue were detected by reverse transcription polymerase chain reaction (RT-PCR).RESULTS: (1) Pt, Po, +dT/dtmax and -dT/dtmax in rat diaphragm in DM group were significantly lower than those in CON group (P<0.05), which were higher in GSH group than those in DM group (P<0.05). (2) Diaphragm MDA content in DM group was increased significantly (P<0.05 vs CON), and SOD activity was significantly decreased (P<0.01 vs CON group); and these changes could be reversed by GSH (P<0.05 ). (3) The levels of nebulin and titin mRNA in DM group were significantly decreased (P<0.01 vs CON group), but increased in GSH group (P<0.05 vs DM group).CONCLUSION: GSH can increase the levels of nebulin and titin mRNA in diaphragm and improve diaphragm contractile force, which may be due to reduce oxidative stress injury.
