Effects of heparin on expression of leptin and transforming growth factor β1 in rat hepatic stellate cells

Abstract: AIM: To explore the effects of heparin on the expression of leptin and transforming growth factor β1 (TGF β1) in rat hepatic stellate cells(HSCs). METHODS: HSCs were divided as follows:heparin group Ⅰ(10 μg/mL), Ⅱ(100 μg/mL), Ⅲ(1 000 μg/mL), and control group.The incubation liquid was extracted after cultured 48 h and frosted in-20 ℃.Leptin and TGF β1 were detected by ELISA, the HSC proliferation was measured by MTT assay. RESULTS: The levels of leptin in heparin group Ⅱ, Ⅲ were significantly lower than that in control group(P<0.01).The level of TGF β1 and the mean absorbance in heparin groups were significantly lower than that in control group (P<0.01). CONCLUSION: Heparin can decrease expression of leptin and TGF β1 in HSCs and inhibit proliferation of HSCs.
