    Effects of crocetin on proliferation and collagen synthesis of hepatic stellate cell

    AIM: To investigate the effects of crocetin on proliferation, collagen synthesis in hepatic stellate cell (HSC) stimulated by acetaldehyde and its partial mechanism.METHODS: HSC stimulated by acetaldehyde was incubated with different doses of crocetin(10-8,10-7,10-6 mol/L), cell proliferation was analyzed by MTT assay, apoptosis ratio was analyzed by flow cytometry(FCM), synthesis of collagen was observed by hydroxyproline concentration measurement. The ERK1/2, Bax, Bcl-2 were tested by Western blotting. Matrix metalloproteinase(MMP-2),the tissue inhibitor metalloproteinase (TIMP-1) mRNA, collagen mRNA were examined by RT-PCR.RESULTS: Within a concentration coverage, crocetin inhibited proliferation of HSC induced by acetaldehyde. The apoptosis rate of cells in crocetin treated groups increased with concentration compared with acetaldehyde group. Crocetin decreased the levels of ERK1/2 and Bcl-2 expression, but increased Bax protein expression. Crocetin could markedly suppressed the up-regulation of CollagenⅠmRNA, Collagen ⅢmRNA, TIMP-1 mRNA, while increased MMP-2 mRNA expression.CONCLUSION: Crocetin inhibited HSC proliferation and collagen synthesis stimulated by acetaldehyde, which related to the expression of ERK1/2 and the regulation of extracellular matrix.

    AIM: To explain the enhancement of bile acid excretion in diabetes from bile acid transporter perspective.METHODS: Primary rat hepatocytes were isolated and cultured in a sandwich configuration (SCRH). Then hepatocytes were cultured with different concentrations of glucose and insulin, and function of Na+/bile acid cotransporter (ntcp) and bile salt export pump (bsep) were tested with Cholyl-lysyl-fluorescein (CLF) as a typical substrate. mRNA levels of ntcp, bsep and lipid regulating nuclear receptor fxr were determined with Real Time PCR.RESULTS: CLF biliary excretion was both decreased in high concentrations of glucose and insulin significantly.Both high concentrations of glucose and insulin can decrease ntcp mRNA for 3-4 fold, but increase bsep mRNA for approximately 2 fold. Levels of fxr mRNA was down regulated in high concentrations of glucose and up regulated in high concentrations of insulin.CONCLUSION: Abnormal glucose and insulin concentration have impacts on bile acid transporters in sandwich cultured rat hepatocytes.Sandwich cultured rat hepatocytes could offer a useful tool to explore hepatobiliary disposition of bile acid and drugs.

    AIM: To construct the recombinant adeno-associated virus (rAAV) vector expressing hS100A1 gene and to verify the correct recombination.METHODS: The hS100A1 plasmid (pUC57-Simple-S100A1) and AAV framework plasmid (pAAV-IRES-ZsGreen1) were cleaved by restriction endonuclease BamHⅠand EcoRⅠ. The target gene fragments were connected together to generate a recombinant plasmid pAAV-IRES-ZsGreen1-S100A1 that was transfected into E. coli DH5a. The plasmid was confirmed to be constructed as expectation by enzyme digestion and sequencing. The recombinant plasmid pAAV-IRES-ZsGreen1-S100A1 was co-transfected into AAV-293 cells with pHelper and pAAV-RC for packaging of recombinant AAV. The efficiency of rAAV packaging was monitored under fluorescent microscope and recombinant viral particles were harvested from infected AAV-293 cells. The virus titer was measured through infecting HEK-293 cells, and the recombinant rAAV-IRES- ZsGreen1-S100A1 was verified by PCR of the exogenous interest genes of S100A1.RESULTS: The recombinant plasmid pAAV-IRES-ZsGreen1-S100A1 was verified by double digestion and sequencing. Using the AAV helper-free system, ZsGreen1 expression could be observed under fluorescent microscope 72 hours after triple plasmid co-transfection and the system provided a high packing ratio of 95%-100%. The rAAV has a high purity and high titer of (2-3)×107 TU/mL. The recombinant virus was confirmed by PCR of exogenous S100A1 genes.CONCLUSION: Recombinant rAAV-IRES-ZsGreen1-S100A1 was successfully constructed with a high virus titer, which may offer the basement of in vitro and in vivo experiments of hS100A1 for gene therapy of chronic heart failure.

    AIM: To explore a method to develop a stable and repeatable murine model of acute promyelocytic leukemia(APL),which progression could be observed.METHODS: For the model, Nod/SCID mice were irradiated by 60Co γ ray and the level of leukocytes was very low at the fifth day, then Nod/SCID mice were injected with HL60 cells intravenously via tail vein.The percentage of HL60 cells in the peripheral blood was determined by flow cytometry, which was used to observe the distribution change of HL60 cells in the peripheral blood.The development of solid tumors in the superficial parts of body was observed daily, and to confirm the characteristics of tumor spread in the mice with histopathological methods.RESULTS: Day 7 post injection, HL60 cells could be detected in the peripheral blood of Nod/SCID mice. About day 30 post injection, solid tumors were observed through the whole body of mice. About day 40, mouse physical fitness began to decline, and mice began to die.The leukemic infiltration was found in the hematopoietic system and non-hematopoietic system. The results of two trials were coincidence.CONCLUSION: For the model, Nod/SCID mice were irradiated by γ ray from 60Co, injected with HL60 cells intravenously via tail vein in the day 5 post irradiation. The course of disease could be kept in about 40 days, and leukemic progression in the Nod/SCID mice with APL was similar with clinical leukemic progression.The model could be used for anti-leukemia drug screening and pharmacodynamic evaluation of candidate drugs with low bone marrow toxicity.
    Protective effects of puerarin on gentamicin-induced nephrotoxicity in rats

    AIM: To explore the possible protective effects of puerarin (Pue) against gentamicin (GM)-induced nephrotoxicity in rats.METHODS: Male rats were i.p. administrated of Pue (50 mg·kg-1·d-1) or vehicle (0.9% NaCl) for 7 days before and 14 days concurrently with saline or GM (140 mg /kg, i.p.). Body weight, serum levels of creatinine (SCr) and blood ureanitrogen (BUN) and creatinine clearance rate (CCr) were assessed, the level of malondialdehyde (MDA), nitric oxide (NO), the activities of superoxide dismutase (SOD) and endothelial nitric oxide synthase (eNOS) in renal tissue were determined. The renal histopathological changes were observed by hematoxylin-eosin staining.RESULTS: Compared with the control group, rats treated with GM showed decrease in body weight, Ccr, NO and the SOD activity in kidney, and a significant elevation of serum SCr, BUN and the renal MDA contents and severe morphology changes. Pue significantly reversed the changes mentioned above.CONCLUSION: Pue provide marked protective effect against GM-induced nephrotoxicity in rats, which may be related to their antagonistic effects on oxidative stress, and the increment of NO level.

    AIM: To investigate the effect of fenofibrate on the expression of short-chain acyl-CoA dehydrogenase (SCAD), cardiac lipid metabolism and cardiac hypertrophy in spontaneously hypertensive rats (SHR).METHODS: SHR were given fenofibrate for 8 weeks, then systolic pressure and the ratios of left ventricular weight to body weight, free fatty acid in blood serum and cardiac muscle were measured. The expression of PPARα and SCAD in myocardium were investigated by western blot. Enzyme activity of SCAD in myocardium was measured with the anaerobic electron transfer flavoprotein fluorescence reduction assay.RESULTS: Compared with WKY rats, systolic blood pressure and the ratios of left ventricular weight to body weight were markedly elevated in SHR. The expression of PPARα and SCAD in myocardium of SHR were decreased as compared with WKY group. The activity of SCAD in myocardium was lower than that in WKY rats. Free fatty acid in blood serum and cardiac muscle were higher than that in WKY rats. Compared with SHR, fenofibrate decreased the ratios of left ventricular weight to body weight, but had no effect on systolic blood pressure. After treated with fenofibrate, the expression of PPARα and SCAD in myocardium of SHR was increased in associated with lower contents of free fatty acid in blood serum and cardiac muscle.CONCLUSION: Fenofibrate may ameliorate cardiac hypertrophy by increasing the expression of SCAD and improving cardiac energy metabolism in SHR.

    AIM: To investigate the expression of signal transducers and activators of transcription 3(STAT3),vascular endothelial growth factor(VEGF), CyclinD1 and their correlation in the lesions of lichen planus.METHODS: The expression levels of STAT3,VEGF and CyclinD1 in the 23 cases of lesions of lichen planus were detected by the immunohistochemistry,and 12 cases of normal human tissues were also detected as control group.The expression and their relationship with pathological factors were analyzed.RESULTS: The expression rates of STAT3,VEGF and CyclinD1 in lichen planus were 60.87%(14/23),69.57%(16/23)and 56.52%(13/23),the expression rates of STAT3,VEGF and CyclinD1 in the normal group were 8.33%(1/12),16.67%(2/12)and 8.33%(1/12).Compared with the control, the difference were significant (P<0.05). Positive correlation between expression of STAT3 and VEGF, STAT3 and CyclinD1, VEGF and CyclinD1(r=0.631, 0.734, 0.564; P=0.001,0.000,0.005) were found.CONCLUSION: STAT3,VEGF and CyclinD1 may play a role in the development in the incidence of lichen planus,coordination between them may play a role.
    Expression of TRPC in the smooth muscle cells of the small intramyocardial arteries in the left ventricles of spontaneously hypertensive rats

    AIM: To investigate the relationship between the remodelling of small intramyocardial arteries(SIMAs) and the expression of transient receptor potential channel (TRPC) in spontaneously hypertensive rats (SHR).METHODS: 24-weeks-old male of SHR taken as the experimental group (SHR group,n=8 ), and the wistar-kyoto rats (WKY,n=8) with matching age and male systolic blood pressure (SBP) is normal as the control group. Left ventriclar mass index (LVMI) was calculated to evaluate the hypertrophy degree of the left ventricle. The structure feature of the small intramyocardial arteries with diameters from 10 to 69 μm in the left ventricles was evaluated with HE staining.The wall area (WA), lumen area (LA), wall area/lumen area (WA/LA) of SIMAs were measured. Vascular smooth muscle cells (VSMCs) were recognized with alpha-smooth muscle actin (α-SMA) immunofluorescence staining.The expression of TRPC1, TRPC3, TRPC5, TRPC6 were also detected with immunofluorescence staining.RESULTS: Both SBP and LVMI of the SHR group were significantly higher than those in WKY group 〔(200±4) vs (118±9) mm Hg;(3.11±0.03) vs (2.42±0.10) mg/g respectively,both P<0.05〕. Both WA and WA/LA of the SHR group were larger than those in WKY group〔(2951±224) vs (2654±190) μm2;(2.01±0.04) vs (1.53±0.03),both P﹤0.05 〕. However, LA of the SHR group was smaller than that of WKY group〔(1469±117) vs (1730±98) μm2 ,P<0.05〕. Positive α-SMA immunofluorescence staining indicated that the thickening middle layer of the SIMAs was smooth muscle cells. The expressions of TRPC1 and TRPC3 on the SIMAs of SHR group were significantly higher than those in WKY group(65±8 vs 28±3 and 60±4 vs 26±8 respectively,both P<0.05). The expressions of TRPC5 in two groups were not statistically different(28±7 vs 28±7,P>0.05). TRPC6 was not expressed on the SIMAs in both groups.CONCLUSION: The smooth muscle cells of the small intramyocardial arteries in 24-weeks-old SHR expresses TRPC1,TRPC3 and TRPC5 but not TRPC6. The remodelling of the small intramyocardial arteries in SHR is associated with the up-regulation of TRPC1 and TRPC3.
    Benzalkonium chloride solution feasibility research on infected abdomen for disposable peritoneal irrigation

    AIM: To observe the antibiosis effects and safety that Benzalkonium chloride (BAC) solution be used in infected abdomen for disposable irrigation.METHODS: To evaluate the antibiosis effects of BAC solution on Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus, Bacillus subtilis, Candida albicans and Aspergillus niger in vitro and the safety for SD (Sprague Dawley) rats and beagle dogs after obtaining two concentrations of BAC solution by disposable peritoneal irrigation. The minimum inhibitory concentrations (MICs) of BAC solution on the cultures were observed by liquid double dilution method that 0.1% original solution was diluted into 15 levels in vitro antibiosis research.The animals were divided into control group, model group and two administration groups which concentrations were 0.005% and 0.01% in the safety research; the route of administration was disposable peritoneal irrigation; irrigating volume was 180 mL/kg body weight for rats and 53.33 mL/kg body weight for dogs; observe the animals' general behavior after the administration, on the eighth day, inspect the general changes of abdominal cavity and the organs in it, then draw blood to detect indexes of blood routine and the histopathology changes of liver, spleen, intestine and peritoneum.RESULTS: The MICs of BAC solution on Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus, Bacillus subtilis, Candida albicans and Aspergillusniger were separately (6.25×10-3)%、(3.12×10-3)%、(9.76×10-5)%、(9.76×10-5)%、(3.91×10-4)% and (6.25×10-3)%. During a week after irrigation, there was no significant difference on general situation; the animals had no obvious visible abnormality in abdominal cavity organs; there was no significant influence on blood routine indexes,no significant histopathology changes on the liver, spleen, intestine and peritoneum in the animals.CONCLUSION: The antibiosis effects of BAC solution on Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus, Bacillus subtilis, Candida albicans and Aspergillus niger in vitro are very good; and it is safe for SD rats and Beagle dogs with below 0.01% (including 0.01%) BAC solution for irrigation of peritoneal cavity only one time. It is feasible that BAC solution is used on infected abdomen for disposable peritoneal irrigation.

    AIM: To study a reasonable algorithm of terminal half-life time (t1/2z) in pharmacokinetics by the accuracy and precision.METHODS: The plasma concentration-time data of pu- erarin was simulated for 1000 subjects by using parameters of intravenous two-compartment models in the variations of 15%, 25% and 35%. Resampling 15, 25, 35, and 45 from the 1000 subjects were used to calculate t1/2z by 4-point, 5-point, 6-point, all-point methods when there were different points in eliminate phase. The results of these methods were compared to the true value and expressed by accuracy and precision.RESULTS: The 5-point method can provide a t1/2z with better accuracy and precision when there are four concentration points in eliminate phase, the 4-point method provide the one with better accuracy in three points in eliminate phase, 6-point method provide t1/2z with better precision in both three and four points in eliminate phase. There is no reasonable t1/2z in all four methods if there are two points in eliminate phase.CONCLUSION: Three concentration points should be at least included in eliminating phase for calculating a t1/2z which is usually more reasonable in 4-points or 5-points method.

    AIM: To investigate the effect of Multienzyme tables digestive vitality in artificial gastric juice because of Belladonna mixture caus.METHODS: Accordance with the compatibility proportion of Belladonna mixture with Multienzyme in artificial gastric juice divided into 3 groups, group A (artificial gastric juice 30 mL),group B (Multienzyme test solution 30 mL+artificial gastric juice 30 mL), group C (Belladonna mixture 30 mL+ Multienzyme test solution 30 mL+artificial gastric juice 30 mL), group B was the control group. Another formulated containing 10, 20, 30,40 mL Belladonna mixture's compatibility liquid was 4 copies. Using Mett glass capillary method to determined digestion vitality of the number of units about compatibility of Belladonna mixture with Multienzyme tables and its compatibility proportion changes in artificial gastric juice.RESULTS: About digestion vigor number of units, group A (120 ± 15) was lower than that in group B (190 ± 20) and group C (104±8) was lower than that in group B(190±20)(P<0.01). Digestive dynamic changes was related to the dose of belladonna mixture(r=-0.9987,P<0.01),also artificial gastric juice's pH values was increased more.CONCLUSION: Multienzyme tables digestion vitality has been a relatively strong inhibitory effect because of Belladonna mixture cause.

    AIM: To observe the change of female estrogens on effects and side effects of sufentanil for postoperative intravenous analgesia after transabdominal hysteromyomectomy(TAH).METHODS: Sixty patients for TAH,ASA I-II ,were divided into two groups:I (premenopansal group)and Ⅱ(postmenopausal group), with 30 cases each. Sufentanil 100 mg and ondanstrone 8 mg were diluted with saline into 100 mL and continuously infused at the rate of 2 mL/h before 30 min end of operation.Visual analogue scale(VAS),Ramsay sedation scale and side effects were recorded at 4,8, 12,24,36,and 48 h postoperatively.RESULTS: Two groups had no significant difference in analgesic effects and sedation scale,VAS of all groups were below 3, sedation scale of two groups were lower than 4.The incidence of side effects -respiratory depression,nausea,vomiting and pruritus were no significant difference in the two groups.CONCLUSION: The change of female estrogens has no significant effects on sufentanil postoperative intravenous analgesia after TAH. Intravenous sufentanil can produce reliable postoperative analgesia for patients with TAH and with fewer adverse effects.

    AIM: To research the effect of oxygen atomization for postoperative cough, expectoration and alleviate the lung infection, and to provide reference for recovery treatment.METHODS: 104 patients with cough were randomly divided into two groups (n=52)in our hospital within one year.The study group was received oxygen atomization treatment,and the control group was received ultrasonic nebulization .RESULTS: The recovery of study group of patients' cough, expectoration and alleviate lung infection recovery were better and faster than those of control group, the difference was statistically significant(P<0.05).At the same time, the complication in study group was less than that of control group (P<0.05).CONCLUSION: Oxygen atomization helps to protect patients' respiratory tract with postoperative.It helps patients sputum expectoration, and also relieves cough and reduces the incisional pain for patients, and it is worthy of popularization and application in clinic.

    AIM: To detect the level of serum γ-glutamyltransferase and evaluate it's relationship with left ventricular diastolic function in type 2 diabetes mellitus.METHODS: A cross-sectional study of 46 type 2 diabetes patients with or without left ventricular diastolic dysfunction were divided into two groups.The levels of serum γ-glutamyltransferase,TC and TG were analyzed.Left ventricular diastolic function was assessed by pulsed Doppler tissue imaging and was determined by ratio E wave/A wave.RESULTS: There were no significant difference about sex, BMI,systolic blood pressure, diastolic blood pressure, HbA1c,total cholesterol and triglyceride in two groups of patients(P>0.05). Compared with the left ventricular diastolic function group,the level of serum γ-glutamyltransferase was obviously higher in left ventricular diastolic dysfunction group,there was significant difference(P<0.01). Negative correlations were found between γ-glutamyltransferase and E/A in left ventricular diastolic dysfunction group(r=-0.681,P<0.000).CONCLUSION: The level of serum γ-glutamyltransferase was increased in type 2 diabetic patients with left ventricular diastolic dysfunction.The level of serum γ-glutamyltransferase might be an marker of diastolic dysfunction in type 2 diabetes patients.

    AIM: To observe the clinical efficacy and security of vinorelbine combined with capecitabine (NX) and gemcitabine combined with cisplatin(GP) for recurrent metastatic breast cancer after anthracycline- and taxane- treatment (ATRMBC).METHODS: 41 ATRMBC patients were divided into NX group(18 cases) and GP group(23 cases).NX group:capecitabine 2000 mg/m2 orally days 1-14 and vinorelbine 25 mg/m2 intravenously days 1 and 8,21 days was a cycle.GP group:cisplatin 20 mg/m2 intravenously days 2 through 5 and gemcitabine intravenously 1000 mg/m2 on days 1 and 8,21 days was a cycle.The dose was adjusted according to adverse effects.The efficacy was evaluated every 2 cycles.RESULTS: All patients were eval- uated for efficacy and toxicity. NX group:one of the 18 patients (5.5%) showed complete response, while 9 patients (50%) showed partial response; the objective response rate was 55.5%. Stable and progressive disease was observed in 4(22.2%) and 4 patients (22.2%), respectively.The median time to progression was 6.3 months;GP group:an objective response was achieved in 13 patients (56.5%). A complete response was achieved in 2 patients (8.9%); partial response in 11 patients (47.8%);stable disease in 5 patients (21.7%), and progressive disease was observed in 5 patients (21.7%).The median time to progression was 6.5 months.There was no significant difference between both groups(P>0.05).The common toxicities included myelosuppression,gastrointestinal reactions,hand-foot syndrome,phlebitis.The incidence rates of platelet decreased and the incidence rate of gastrointestinal reactions in GP group were higher than those in NX group, and there were differences between the two groups (P<0.05),the incidence rate of hand-foot syndrome in NX group was higer than that in GP group, and there were significant differences between the two groups(P<0.01).CONCLUSION: Vinorelbine combined with capecitabine (NX) and gemcitabine combined with cisplatin(GP) are effective for patients with ATRMBC.The hematologic and non-hematologic toxicities are well tolerated.

    AIM: To investigate the ratio of prophylactic antibiotics in three kinds of type I incision operation.METHODS: Each 120 cases including thyroid surgery, breast surgery, inguinal herniorrhaphy from September to December in 2011 were analyzed.RESULTS: In the 360 cases, the ratio of antibiotics was 75.8%, the ratio of indication of prophylactic was 31.9%, the indication of prophylactic use timing is reasonable accounted for 71.4%.The main volume of solvent was 250 mL.CONCLUSION: There are main problems that the indication of prophylactic is low, the volume of solvent is too large in prophylactic antibiotics use in three kinds of type I incision operation. Some necessary intervention should be taken in order to promote the rational use of antibiotics.

    The epithelial-mesenchymal transition (EMT) plays important roles in many physiologic and pathological progresses. Recent researches showed that EMT involved in the liver fibrosis. In this review, we first describe the cells in the liver which undergo EMT, including hepatocytes, cholangiocytes, hepatic progenitors and hepatic stellate cells. Then, we systematically present the possible signal pathways and regulatory mechanisms of EMT in liver fibrosis. Finally, the feasibility of anti-fibrosis treatment targeting EMT is discussed as well.

    Dihydropyridines represent a class of the most important calcium antagonists which are widely used as antihypertensive drugs. In this review, we summarized the published metabolic and pharmacokinetic studies of dihydropyridines, including their absorption, distribution, metabolism and excretion in animals and humans, to provide reference for drug development and clinical application. Dihydropyridines are highly lipophilic with good membrane permeability, but subjected to extensive first-pass effect which affects their oral bioavailability; these drugs, with large apparent volume of distribution and high plasma protein binding rate, can rapidly distribute to various tissues, and the fast blood flow rate tissues such as liver and kidney are the main distribution tissues; they are metabolized mainly by cytochrome P450 enzymes (CYPs), especially CYP3A isoforms.Oxidation of the 1,4-dihydropyridine into pyridine, ester hydrolysis and hydroxylation are involved in phase I metabolism,the phase II metabolism is glucuronidation; they are excreted from urine, bile and feces mainly in the form of metabolites, the excretion of parent drug is little.

    Follicle stimulating hormone (FSH) is a glycoprotein hormone played an essential role in reproduction and widely used in the treatment of infertility. However, in the assisted reproductive technology programs, the response of ovulating women to exogenous FSH is quite variable in individuals. FSH action is mediated by the FSH receptor (FSHR). More than 900 polymorphisms of FSHR gene are known, it has been previously reported that SNPs of FSHR may be related to the response of the patients to FSH stimulation and should, therefore, be considered in assessment of efficacy in controlled ovarian hyperstimulation during assisted reproduction techniques in women. The aim of these studies would be the prediction of the genetic causes of diseases to enable an individual clinical setting according to the patient's genetic background undergoing FSH stimulation. In this review we discuss the latest information about the effects of polymorphisms of the FSHR genes on reproductive functions of female, and discuss their clinical implications.

    Diabetes is a metabolic disorder syndrome characterized by hyperglycemia and cardiovascular complications are major causes of death and disability. 20-hydroxyeicosatetraenoic acid (20-HETE) is the newly-identified active metabolite of arachidonic acid (AA) catalyzed by cytochrome P-450 (CYP450), which is called as the third metabolic pathway of AA. Recent researches suggest its significant role in the regulation of renal function and vascular smooth muscle tension. But there is few reports about the relationship of 20-HETE and diabetes. The present article reviewed the international researches on 20-HETE and diabetic cardiovascular complications published in recent years.
