Effect of recombinant human keratinocyte growth factor-2 on burn wound healing in rats and its probable mechanisms of action

Abstract: AIM:To investigate the effect of recombinant human keratinocyte growth factor-2 (rhKGF-2) on skin wound healing in rats and its possible mechanism.METHODS:Deep Ⅱ degree burn wound model was established in rats. The wound areas, the thickness, migration distances and areas of neoepithelia tissue were detected with IMAGE-PRO express morphological analysis software after Hematoxylin and Eosin staining. MTT and clone forming assay were used to measure the HaCaT cells proliferation. The keratinocyte migration and cell wound closure were observed in the scratch experiment.RESULTS: rhKGF-2 obviously accelerated the healing rate of deep II degree burn wound with improved reepidermidation. Increases in migration distance, average area and thickness of neoepithelium were found in a dose-dependent manner. In addition, rhKGF-2 increased HaCaT cells proliferation rate, migration and clone forming efficiency in a concentration dependent manner (P<0. 05).CONCLUSION: These results show that rhKGF-2 significantly accelerates wound healing partly through the enhancement of keratinocyte proliferation and migration.
