    AIM: To study the effects of 6-OHDA and MPP+on the glutamate uptake into cultured C6 glioma cells.METHODS: The glutamate uptake into C6 glioma cells was measured by using radioimmunoassay method;and the viability of C6 glioma cells was investigated by MTT method.RESULTS: Both 6-OHDA and MPP+inhibited glutamate uptake into C6 glioma cells in a concentration-dependent manner.Treatment with 6-OHDA produced inhibitory effect on the viability of C6 glioma cells.However,exposure to MPP+showed no such effect.Incubation with TPA,a PKC activator,caused increase in glutamate uptake and completely reversed MPP+-induced uptake inhibition.CONCLUSION: Inhibition of glutamate uptake by MPP+was completely reversed by TPA,indicating that the effect of MPP+was mediated by PKC cell signal transduction,while the inhibitory effect of 6-OHDA was due to the impairment viability of C6 glioma cells.

    AIM: To identify the function ORF(open reading frame)U41 of human herpesvirus 7(HHV-7).METHODS: Immunofluorescent detection was used to show the localization of the U41 protein in cells.The expression kinetics of the U41 gene in the presence of cycloheximide(CHX)and phosphomoformic acid(PFA),which are inhibitors of protein synthesis and of viral DNA synthesis respectively,was detected with RT-PCR.DNA binding assay was used to detect the single and double DNA strands binding ability of U41 protein.RESULTS: The U41 protein was expressed as an early protein in the HHV-7 infected cells and localized only in the nuclei of infected cells.The U41 protein was able to bind to a single-strand DNA cellulose column but not to double-strand DNA cellulose column from DNA binding assay.CONCLUSION: The major function of U41 may be to keepthe DNA template in the optimal conformation for DNA elongation and for viral DNA synthesis.

    AIM: To observe the improvement function of huperzine-A transdermal drug delivery system(Hup-A TDDS)administration at Governor Vessel(GV)acu-points on memory disorders induced by scopolamine in mice and to compare the effect of GV acu-points administration with general place administration.METHODS: Stepdown test and “Y” maze test were used to observe the influence of Hup-A TDDS administration at GV acu-points on memory disorders induced by scopolamine.Some biochemical indices were also etermined.RESULTS: (1)The marked effects were found among 3 -6 h after Hup-A TDDS administration at GV baihui acupoint.The best effects showed 4 h after administration,and it weakened a little from 5 h to 6 h in 3.4 μg/0.25cm2 group.At 26.7 μg/0.25 cm2 group,the drug action maintained an effective level from 3 h to 6 h,and reached the maximum at 4 h.(2)Among 5 acu-points of GV to administration at Hup-A TDDS 26.7μg/0.25cm2,the obvious effects on memory disorders existed,and baihui(GV20)was the most effective acu-point.(3)The administration of Hup-A TDDS(26.7μg/0.25cm2 and13.4 μg/0.25cm2)through both acu-point and general place(buttocks),approved memory improvement function by stepdown test and “Y” maze test,and the action in baihui acu-point groupshowed more significant.The depressed trend of cholinesterase activity in brain was proved in all of both GV acu-point and general place groups and baihui acr-point groupexposed signal difference when compared with mokcl group.CONCLUSION: Hup-A TDDS administration at GV acu-point shows a better improvement of memory disorders in comparison with general place.The action mechanism may be due to the depressed cholinesterase activity and the improved cholinergic system function.

    AIM: To study the effects of sodium magnesium fructose diphosphate(FDPM)on lactate concentration and ATPase activity of ischemic synaptosome,so as to explore the protective mechanisms of FDPM on cerebral ischemia.METHODS: Synaptosome prepared from normal rat brain was cultured with oxygen and glucose deprived to establish the model of ischemic synaptosome.1.3 mmol·L-1magnesium sulfate,4.0mmol·L-1 fructose-1,6-diphosphate(FDP)and 1.3 mmol ·L-1 FDPM were incubated with the ischemic synaptosomes separately.After 60min,the lactate concentration as well as the activities of Na+-K+ATPase and Ca2+-Mg2+ATPase were detected.RESULTS: FDPM reduced the lactate concentration,raised the activities of Na+-K+ATPase and Ca2+-Mg2+ATPase of ischemic synaptosomes(p<0.05).The effects of FDPM were stronger than those of FDp(p<0.05).CONCLUSION: FDPM may protect cerebral ischemia by improving the energy metabolism,and mitigating acidosis state of brain tissue.

    AIM: To explore the difference of efficacy between defibrase and other drugs in treatment of acute cerebral infarction.METHODS: The efficacies of defibrase or other drugs in treatment of acute cerebral infarction were analyzed by Meta-analysis in homogeneity test and combined test in 10 studies.RESULTS: (1)Homogeneity test showed that the cited studies were homogeneous with χ2=6.8452,p>0.05;and(2)in combined test,the combined OR=3.2353 and its 95% confidence interval was in 2.21 -4.74(χ2=36.2,p<0.001).CONCLUSION: The effectiveness of defibrase in treatment of acute cerebral infarction is significantly superior to those of the medications used at present.It is necessary to further investigate the relationships between its clinical efficacy and the dosages,and between protocols of drug administration and indications.

    AIM: To study the pharmacokinetics and relative bioavailability of propylthiouracil tablet in Chinese healthy male volunteers.METHODS: The bioequivalence and relative bioavailability of a test domestic formulation of propylthiouracil in comparison with a reference imported formulation was investigated in a randomized 2-way cross-over study in 20 healthy volunteers following a single oral dose of 200 mg.The washout period was 1wk.Plasma concentrations of propylthiouracil were determined by a HPLC system equipped with a ultraviolet detector.RESULTS: The following pharmacokinetical parameters were derived from the individual plasma concentration time courses.The main parameters for test and reference formulations,Cmax were(5.7±1.5)and(5.8±1.8)mg·L-1;tmax were(1.4±0.8)and(1.4±0.7)h;t1/2were 1.7±0.3 and(1.7±0.2)h;AUC0-t were(18.0±4.4)and(17.8±4.6)mg·h·L-1;AUC0-∞were(18.1±4.5)and(17.9±4.7)mg·h·L-1,respectively.Compared with reference formulation,the relative bioavailability(F)of test formulation was(101.9±13.2)%。The above parameters obtained from two formulations were compared by analysis of variance(ANOVA)and no significant difference was found.The bioequivalence of two formulations was found assessed by two one-side ways test(p<0.05).CONCLUSION: Therefore,from the results of this study it can be concluded that the test formulation is bioequivalent with the referenceformulation.

    AIM: To compare the effects of three types of antihypertensive drugs of delayed-action preparation(DAP)and three types of typical preparation(TP)on the blood pressure of patients with subcortical arteriosclerotic encephalopaphy.METHODS: Sixty-six patients with subcortical arteriosclerotic encephalopaphy were randomly divided into DApgroupand Tpgroup,each groupincluding 33 patients.Three types of antihypertensive drugs of DApand TP,including calcium antagonist,β-receptor blocker and angiotensin converting enzyme inhibitor,were used orally in respective groupfor ten consecutive weeks.Blood pressure and heart ratewere monitored dynamically.Activities of daily living(ADL)of each patient were investigated before and after treatment.RESULTS: Total effective rates of antihypertension were 93.9% and 87.9% in DApgroupand in Tpgroup,respectively(p>0.05).The effects of DApwere found to be stable in day time and at night.The rates of side effects were 15.2% and 42.4%(p<0.05)in DApgroupand Tpgroup,respectively.Most side effects were mild and transient in DApgroup,without significant effect on the heart rate.There was not significant change on ADL scores in both groups after the treatment(p>0.05).CONCLUSION: Antihypertensive drugs of ADpare effective agents with a good tolerance in treatment of hypertensive patients with subcortical arteriosclerotic encephalopathy.

    AIM: To investigate the effect of propofol on myocardial malondialdehyde(MDA)metabolism and water content after ischemia/reperfusion injury in isolated rat heart with the modified Langredorff model.METHODS: Twenty four rats were randomly divided into four groups.Rat hearts were perfused with Krebs-Henseleik(K-H)in normal control group.In other three groups,a three-phase protocal was performed:(1)15 -30 min preperfusion,(2)20 min global normothermic(37℃)ischemia,and(3)30 min reperfusion.Then the grouppwas treated with 30 μmol·L-1propofol,the groupp+G 30 μmol·L-1 propofol+10 μmol·L-1 glibenclamide before ischemia and throughout the experiment.Only K-H buffer was perfused in the groupI-R.The content of water and MDA in myocardium and creatin kinase(CK)in the coronary effluent were measured.RESULTS: The recovery of hearts treated with propofol were better than that of groupI-R,indicated by lower content of water,MDA and CK level(p<0.01).No statistically significant differences were observed between grouppand groupp+G(p>0.05).CONCLUSION: Propofol can relieve myocardial edema by scavenging oxygen free radical and the mechanism is related to open of KATpchannels.

    AIM: To study the antitumor effect of the crude or the refined extract of glycosaminoglycan(CGAG or GAG)from whole viscera of pinctada martensis.METHODS: The antitumor actions of CGAG or GAG were examined in KM mice inoculated S180 and EAC,including inhibitory rate of tumor growth,the ratio of extending viability,the synergism with 5-FU or Tegafur.The inhibition to the growth of HL-60 cells was observed in vitro.RESULTS: GAG reinforced the antitumor effect of 5-FU or Tegafur in S180 or EAC mice,and the inhibitory rates of tumor growth were 61.96%(S180)and 46.30%(EAC).GAG lengthened life time by 14.16% on S180 mice.The kill rates to HL-60 cells in vitro were 38.67% and 25.64% by using the CGAG(1.0 or 0.1g·L-1).CONCLUSION: GAG can increase the antitumor effects of 5-FU and Tegafur in S180 and EAC mice,lengthen life time in S180 mice,and kill the HL-60 cells in vitro.

    AIM: To observe the cytotoxicity effect of scorpion venom crude(SVC-Ⅲ)on the human jugular squamous cell carcinoma(AGZY-973)and to find a properly consistence of SVC of the growth inhibiting to AGZY-973.METHODS: MTT colorimetric method was used to study the growth inhibition rate(GIR)in different condition.RESULTS: The ability to metabolize MTT of AGZY-973 cells was lower than control distinctly after the cells were treated by SVC-Ⅲ.The growth of AGZY-973 cells was inhibited obviously by SVC Ⅲ and GIR was reached 68.96%(p<0.01).The dose-response relationshipwas unclear when consistence were more than 40mg·L-1,while the dose-response relationshipwas clear when dose was in 10 -40 mg·L-1.The growth inhibition rate(GIR)of 5-Fu reached 55.27%(p<0.01).CONCLUSION: SVC is one kind of effective antineoplastic components in the venom of buthus martensii scorpion.It possesses growth inhibition on AGZY-973 cells.The properly consistence of SVC of the growth inhibition to AGZY-973 cells probably is 40 mg·L-1.

    AIM: To explore in vitro the effects of WR-2721,a cytoprotector of chemotherapy and radiation,on platelet activation induced by physiologic agonists ADP,collagen and PAF.METHODS: Blood,obtained from Chinese healthy volunteers aged 20 -35 years,was treated with physiologic agonists at the given concentrations:ADp1 μmol·L-1,collagen 2 mg·L-1,and PAF0.1 mg·L-1.Then WR-2721 was added to the experimental system at final concentrations range of 10-7 -10-5 mol·L-1.The effect of WR-2721 on platelet aggregation induced by the agonists was studied.Thromboxane B2(TXB2)levels and nitric oxide(NO)production were determined.RESULTS: WR-2721 inhibited both platelet aggregation and TXB2 production induced by ADP,collagen and PAF,in a dose-dependent manner.When WR-2721 was added at the final concentration of 5μmol·L-1,it significantly increased ADP,collagen and PAF-induced NO production,which suggested that NO release by activated platelets was involved in the inhibitory effect of WR-2721.CONCLUSION: WR-2721 can effectively inhibit platelet activation induced in vitro by physiologic agonists.It may be not only a cytoprotectant,but also a therapeutic agent.

    AIM: To observe platelet activation and fibrinolytic activity and evalute the effects of valsartan and amlodipine in elderly patients with essential hypertension.METHODS: Double antibody sandwich ELISA and spectrophotometric assay were used to examine the levels of platelet alpha-granule membrane protein(GMP-140),tissue-type plasminogen activator(t-PA)and its inhibitor(PAI-1)in 57 elderly patients with essential hypertension and 30 normotensives.Valsartan was given in 29 cases(groupⅠ)and amlodipine in 28 cases(groupⅡ)for 12 weeks.RESULTS: Elevated GMP-140,decreased t-PA activity,and elevated PAI-1 activity were detected in the patients,but not in normotensives(p<0.01).Levels of GMP-140 were positively correlated with PAI-1 in the patients(p<0.01).GMP-140 and PAI-1 activity were decreased(p<0.01 -0.05),but t-PA activity was increased(p<0.05)after treatment in both group.No significant difference was found in therapeutic effect between groupⅠ and groupⅡ.CONCLUSION: In elderly patients with essential hypertention,the platelet activation elevates and fibrinolytic activity decreases,and both valsartan and amlodipine can inhibit platelet activation and improve fibrinolytic activity.

    AIM: To investigate the pharmacokinetics and relative bioavailability of shuanghuanglian injection in rabbits by rectal medication in comparison with oral medication.METHODS: Chlorogenic aci from flos loniceraed,a main component in shuanghuanglian injection,was used as a detection marker in blood drug.According to the velocity of drug entering into systemic circulation from medication site(that is,absorption rate Ka)and the relative quantity of drug entering into systemic circulation from medication site(that is,absorption degree),AUC was usually used as a marker to observe absorption dynamic changes of rectal medication.RESULTS: There was significant difference of AUC between clusis groupand oral group(p<0.05).Bioavailability of enema was 100%,and oral dose 80.87%;The values of Tp of enema and oral dose were 0.75 h and 2.48 h,respectively(p<0.05),showing that clusis was superior to oral medication;Cmax was affected by absorption quantity and time.There was significant difference of Cmax between enema and oral dose(p<0.05),showing that the blood drug concentration of enema was obviously higher than that of oral dose.CONCLUSION: For the chlorogenic acid from shuanghuanglian injection,the absorption in rectal medication is faster than that in oral medication,and the blood concentration,total absorption quantity and bioavailability of rectal medication are higher than ones of oral medication.

    AIM: To observe the effect of L-arginine on blood pressure(BP)in stress rats.METHODS: Thirty rats were divided into three groups.L-ARG +stress group:L-arginine was added in drinking-water and electric stimulus were given intermittently in rat foot-plate for 15 days;stress group:electric stimulus was given as above;control group:no stress.The Bplevels of tail artery were measured per two days,and the concentrations of nitric oxide(NO)of plasma were detected on 15th day.RESULTS: There were significantly differences on Bpbetween control groupand stress groupin the 9th,11th,13th,15th day(p<0.05 or p<0.01).After the stimulus in the stress group,the level of Bpincreased,the concentration of NO decreased(p<0.05).However,the Bpdid not show obvious difference between control groupand L-ARG +stress group.On the 15 th day,the Bplevel decreased in L-ARG +stress group(p<0.05),and the concentration of NO was higher than that of control group(p<0.01).CONCLUSION: L-ARG can resist the increase of Bpin stress rats.

    AIM: To evaluate the efficacy and safety of a combination therapy of clarithromycin(CLA),colloidal bismuth subcitrate(CBS),and metronidazole(Met)in treatment of helicobacter pylori(HP)infection with recurrent abdominal in children.METHODS: By using the 14C-urea breath test(14 C-UBT)and serum HPIgG antibody test,28 patients of Hpinfection with recurrent abdominal pain were divided into treatment groupand control group.The treatment groupwas given CLA 15 mg·kg-1 bid;CBS 8mg·kg-1 bid;Met 18 mg·kg-1 bid for1 wk.The control groupwas administered CLA 15 mg·kg-1 bid;CBS 8 mg·kg-1 bid;amoxicillin 40 mg·kg-1bid for 2 wk.All patients were followed-up4 wk after the treatment was completed and14C-UBT was repeated.RESULTS: Hperadication rates were 92.3% in treatment groupand 86.7% in control group(p>0.05).The pain relief rates were not statistically significant(p>0.05)between two groups.The side effects were 7.7%and 13.3% in treatment groupand control group,respectively.CONCLUSION: The combination of CLA +CBS+Met for 1 wk is an effective and safe treatment for eradication of Hpinfection with recurrent abdominal in children.

    AIM: To observe the effects of glucomannan on nitric oxide(NO),endothelin(ET),lipid peroxide and superoxide dismutase(SOD)in experimental atherosclerotic rabbits.METHODS: An atherosclerotic rabbit model was established by feeding high cholesterol diet supplemented by bovine serum albumin injection bolus.The rabbits were randomly divided into the control,model,and glucomanan treatment group.Blood samples were collected 8 weeks after the treatment.RESULTS: After glucomanan administration for 8 wk,the NO and activity of SOD increased in comparison with model group(p<0.01),while MDA and ET decreased(p<0.01).CONCLUSION: Glucomanan has the effects of antioxidation,preserving endothelial function,and maintaining the balance of NO/ET.

    AIM: To observe the effects and safety of clopidogrel used in percutanous coronary intervention(PCI)while acute occlusions of coronary arteries happened.METHODS: 92 patients with acute occlusions of coronary arteries were treated through PCI and they were randomized equally into following groups:clopidogrel group(CPG group)and ticlopidine group(TCpgroup).Each patient was treated with clopidogrel 25 mg bid or ticlopidine 250 mg bid for 4 weeks after PCI.RESULTS: Compared with the control group,there was not significantly difference in platelet aggregation(45.36±2.98 vs 51.60±2.56)and clinical effects(97.95% vs93.02%)after 3 weeks.The proportion of patients with adverse events who had to release the treatment was2.04%(CPG group)and 16.27%(TCpgroup).CONCLUSION: The clinical effect of clopidogrel is similar to ticlopidine and shows less adverse events.

    AIM: To investigate the effects of losartan on myocardial fibrosis and left ventricular hypertrophy in hypertensive patients.METHODS: 35 essential hypertensive patients with left ventricular hypertrophy were treated with losartan 50 -100 mg for 6 mon.Before and after the treatment,serum procollagen type Ⅲ(PC Ⅲ),laminin(LN),and hyaluronic acid(HA)were measured by radioimmunoassay;and left ventricular mass index(LVMI)was determined with ultrasonic cardiogram.RESULTS: After treatment with losartan:the serum levels of PC Ⅲ,LN,HA,and LVMI were all decreased(p<0.05 or p<0.01),and the blood pressure was also decreased significantly(p< 0.01).CONCLUSION: Losartan can decrease blood pressure,and reverse the left ventricular hypertrophy and myocardial fibrosis in hypertensive patients.

    AIM: To estimate the efficacy of milrnone in treatment of congestive heart failure.METHODS: 122 cases of chronic congestive heart failure with different etiology were divided into milrinone groupand dabutamine grouprandomly.Milrinone groupaccepted milrinone(10mg·d-1)for 5 days,and dabutamine groupaccepted dabutamine(160 mg ·d-1)for 5 days.The efficacy of treatments was evaluated.RESULTS: The effectiveness rates of cardiac function improvement were 93.9% and76.7% in milrinone groupand dabutamine group,respectively(p<0.05).The cardiac function improvement accompanied by the heart rate decrease and stable blood pressure in milrinone group.CONCLUSION: Milrinone is an effective drug for chronic congestive heart failure.

    AIM: To re-evaluate the effects of Beijing Hypotensive No.0 in the treatment of essential hypertension.METHODS: The 39 cases of patients suffered essential hypertension were treated by Beijing Hypotensive NO.0 at 1 tablet each time and once a day.A single course of treatment lasted for eight weeks.The trial was evaluated with blood pressure,heart rate,blood routine examination,hepatic and kidney function,blood sugar,blood lipid,and blood uric acid before and after the treatment.RESULTS: Beijing Hypotensive NO.0 could significantly relieve hypertension(p<0.01);no differences were shown in blood routine examination,hepatic and kidney function,blood lipid,blood sugar,and blood uric acid(p>0.05).CONCLUSION: Beijing hypetensive No.0 is an effective and safe agent in the treatment of essential hypertension.

    AIM: To evaluate the therapeutic effect of artificial liver support system(ALSS)in the treatment of severe hepatitis.METHODS: Seventy patients with severe hepatitis were enrolled in two groups:treatment group(n=32)given ALSS combined with a routine treatment,and control group(n=38)only given the routine treatment.RESULTS: The liver functions of patients in treatment groupwere significantly improved after ALSS therapy.The total clinical efficacy rates in treatment groupand control groupwere 46.9% and 23.2%,respectively(p<0.05).The efficacy rates of patients receiving ALSS therapy in early,middle and late stage were 80.0%,50.0% and 10.0% respectively.CONCLUSION: ALSS is effective in treatment of severe hepatitis,especially in early and middle stage.

    AIM: To establish a HPLC method to determine nifedopine the content and uniformity of nifedopine tablet.METHODS: HPLC condition was set as ODS column,flow phase:methanol-water(60 ∶40),flow rate:0.8 ml·min-1,and detection wavelength:235nm.RESULTS: The nifedopine linear range was 0.020-0.200 g ·L-1 in this method.The average recovery rates were 99.6%,98.9%,and 100.4% in high,normal and low concentrations,respectively.The corresponding values of RSD were 0.42%,0.54% and 0.35%(n=5).CONCLUSION: This HPLC method shows a high specificity and sensitivity,and is accurate,simple and rapid in performance.

    AIM: To explore the immunologic effect of extra bacillus calmette guerin(EBCG)on asthmatic patients.METHODS: 58 asthmatic patients in acute exacerbation were randomly enrolled into two groups:EBCG group(n=34)and control group(n=24).Before treatment,the levels of interleukin-4(IL-4)and interleukin-12(IL-12)in blood plasma were examined in both groups.The patients in EBCG groupinhaled budesonide aerosol according to asthmatic ladder treatment program and injected with EBCG.Spasmolytic could be used if necessary(excluding theophylline).The patients in control grouponly used budesonide aerosol and spasmolytic.The level of IL-4 and IL-12 in blood plasma were determined again after three months.RESULTS: After the drug intervention,the level of IL-4 significantly decreased in EBCG group(29.0±5.6 vs 22.6±6.5 ng·L-1,P<0.05),but IL-12 concentrations in plasma markedly increased(37.2±4.6 vs 51.7±10.7 ng ·L-1,p<0.05).In control group,the level of IL-4 decreased from(29.7±5.1)to(26.4±4.9)ng·L-1(p<0.05),but IL-12 concentrations did not significantly change(p>0.05).Before drug intervention,no significant differences were found between two groups in IL-4 and IL-12concentrations;However,there were markedly differences after drug intervention(p<0.05).CONCLUSION: EBCG can restrain the secretion of IL-4 and contribute to the secretion of IL-12 in asthmatic patients.
